NNMT is highly active in white adipose (fat) tissue, particularly in states of obesity and metabolic dysfunction. When NNMT activity is elevated, it diverts nicotinamide away from the NAD⁺ salvage pathway and consumes methyl donors such as SAM (S-adenosylmethionine). This can influence cellular energy production, mitochondrial function, and epigenetic signalling.

By inhibiting NNMT, 5-Amino-1MQ is thought to:
